Semaphorin profiling of periodontal fibroblasts and osteoblasts.

چکیده
UNLABELLED Cells of the periodontal attachment (cementoblasts, osteoblasts, and periodontal ligament fibroblasts) are descended from a common progenitor (the cranial neural crest). During their differentiation into different cell types, these cells separate from one another to form a laminated structure. Semaphorins (and their neuropilins and plexin receptors) act as cell guidance molecules for other neural crest derivatives. It is predicted that the differential expression of these molecules will correlate with the ability of these cells to segregate. It is demonstrated that human pre-osteoblasts segregate from PDL and gingival fibroblasts in culture. In addition, these cells express different semaphorins and plexins. Semaphorins 3D and 7A were expressed preferentially in dermal fibroblasts, while semaphorin 6B was selectively expressed by pre-osteoblasts. Semaphorins 3B, 4C, 5B, and 6C and plexins B1 and C1 were expressed in reduced levels in pre-osteoblasts. Analysis of the data suggests that differential expression of semaphorins and plexins may be involved in regulating cell-sorting in the formation and regeneration of the periodontal attachment structure. ABBREVIATIONS Periodontal Ligament (PDL), Reverse Transcriptase Polymerase Chain-reaction (RT-PCR).

Comparison of Biocompatibility of Various Membranes with Fibroblasts
Objective: Different techniques have been suggested for the repair of bone defects at the injured sites. Use of biomembranes, or application of plasma rich in growth factor (PRGF) at the site of proliferation of osteoblasts are among the suggested techniques. The current study aimed to compare the biocompatibility of human periodontal ligament fibroblasts (hPLF) cultured on Hypro-Sorb F, Peri...
متن کامل[Study on proliferation and function of periodontal ligament fibroblasts and osteoblastic cells under hypoxia].
There have been many reports recognizing vascular changes on pressure side of periodontal tissues during orthodontic tooth movement. The vascular changes cause local hypoxia which seems to affect the phenotypes of periodontal tissue cells. In order to clarify the effect of hypoxia on proliferation and function of periodontal tissue cells, DNA content, alkaline phosphatase (ALP) activity and pro...
متن کاملIn vitro-activity of oily calcium hydroxide suspension on microorganisms as well as on human alveolar osteoblasts and periodontal ligament fibroblasts
BACKGROUND Findings from animal and human studies have indicated that an oily calcium hydroxide suspension (OCHS) may improve early wound healing in the treatment of periodontitis. Calcium hydroxide as the main component is well known for its antimicrobial activity, however at present the effect of OCHS on the influence of periodontal wound healing/regeneration is still very limited. The purpos...
متن کاملThe Role of Proteinase-Activated Receptors 1 and 2 in the Regulation of Periodontal Tissue Metabolism and Disease
Proteinase-activated receptors 1 (PAR1) and 2 (PAR2) are the most highly expressed members of the PAR family in the periodontium. These receptors regulate periodontal inflammatory and repair processes through their activation by endogenous and bacterial enzymes. PAR1 is expressed by the periodontal cells such as human gingival fibroblasts, gingival epithelial cells, periodontal ligament cells, ...
متن کاملCapturing the Regenerative Potential of Periodontal Ligament Fibroblasts
The cell population within the periodontal ligament (PDL) tissue is remarkably heterogeneous1. Fibroblasts, a mixed population of cells, are the main cellular component of the PDL and the cell type most often studied for periodontal regeneration. Osteoblasts and osteoclasts are found on the bone side, while fibroblasts, macrophages, undifferentiated adult/mesenchymal stem cells, neural elements...
